Neil Woods’s side went a goal down in the first minute as Doncaster came out the traps quickly to make it 1-0.
And the home side doubled their lead inside the opening 10 minutes, before making it 3-0 15 minutes later.
Town, who had a disastrous start, began to get a foothold in the game for the remainder of the first half, but struggled to grab a goal.
But the young Mariners made three changes at the break and continued to plug away as they searched for a way back into the game.
And they got on the scoresheet as Lennon Stead smashed home from close-range, after some neat play from Town.
But Doncaster responded well to conceding and re-established their three-goal lead as they scored a fourth.
Both sides subsequently had multiple goalscoring opportunities, but the game finished 4-1.
The young Mariners are back in action against Bradford City at Cheapside on Saturday (11am).
